Recovery Effect of DA-3030, A Recombinant Human Granulocyte Colony-Stimulating Factor, on Neutropenia Induced by Anticancer Agents in Mice

Abstract
Background:
@EN Neutropenia is a major dose-limiting factor in cancer chemotherapy diminishing its usefulness and increases patient's susceptibility to infectious disease. Some recombinant human granulocyte-colony stimulating factors(rhG-CSFs) are in use to
reduce
the risk of this serious side effect. The present study was designed to evaluate the recovery effects of DA-3030, a rhG-CSF produced in E. coli, and filgrastim (Grasin(r)) on neutropenia induced by anticancer agents in normal and tumor-bearing
mice.
@ES Methods:
@EN Neutropenia in normal mice was induced by a single combined intraperitoneal injection of 130mg/kg of cyclophosphamide(CPA). 4.5mg/kg of doxorubicin, and 1mg/kg of vincristine into ICR mice on day 0. Neutropenia in tumor bearing mice was made
by
a
single intraperitoneal injection of 200mg/kg of CPA into Colon 26 adenocarcinoma-bearing BALB/c mice 7 day after tumor implantation. DA-3030 or filgrastim (100§¶/kg/day) was subcutaneously administered for 5 consecutive days starting 1 day after
injection of anticancer agents. The changes in peripheral blood cells(WBC, RBC, Platelets, Neutrophils) were observed for 2 weeks.
@ES Results:
@EN The neutrophil count in peripheral bloods of ICR and BALB/c mice reached nadir on 3 day and 4 day after chemotherapy, respectively, and a significant decrease in neutrophil number persisted around to 7 day after chemothrapy. Injection of
DA-3030
accelerated the recovery from these anticancer drug-induced neutropenia. There were no significant difference in the RBC counts, Platelet counts and body weight change between control and DA-3030 group. Filgrastim at the same dose produced the
same
effect.
@ES Conclusion:
@EN These results suggest that DA-3030, like filgrastim, could be useful in the clinical treatment for neutropenia induced by anticancer agents.
